Therapie mit Methotrexat

Methotrexat wird in der onkologischen (z.B. akute lymphatische Leukämie, non-Hodgkin-Lymphom) als auch in der immunsuppressiven Therapie (z.B. rheumatoide Arthritis) eingesetzt. Die Einnahme von Methotrexat beeinflusst wesentlich den Folsäurestoffwechsel. Folsäure ist ein wichtiges Spurenelement, das an der Synthese einzelner Nukleotidbausteine beteiligt ist. Die Hemmung des Folsäurestoffwechsels durch Methotrexat birgt neben dem gewünschten antientzündlichen und antiproliferativen Effekt auch das Risiko für das Auftreten unerwünschter Nebenwirkungen. Eine genetische Variante im Gen der Methylentetrahydrofolatreduktase (MTHFR) ist mit einem erhöhten Nebenwirkungsrisiko assoziiert.
